RESUMO

OBJECTIVES@#To construct a prognosis risk model based on long noncoding RNAs (lncRNAs) related to cuproptosis and to evaluate its application in assessing prognosis risk of bladder cancer patients.@*METHODS@#RNA sequence data and clinical data of bladder cancer patients were downloaded from the Cancer Genome Atlas database. The correlation between lncRNAs related to cuproptosis and bladder cancer prognosis was analyzed with Pearson correlation analysis, univariate Cox regression, Lasso regression, and multivariate Cox regression. Then a cuproptosis-related lncRNA prognostic risk scoring equation was constructed. Patients were divided into high-risk and low-risk groups based on the median risk score, and the immune cell abundance between the two groups were compared. The accuracy of the risk scoring equation was evaluated using Kaplan-Meier survival curves, and the application of the risk scoring equation in predicting 1, 3 and 5-year survival rates was evaluated using receiver operating characteristic (ROC) curves. Univariate and multivariate Cox regression were used to screen for prognostic factors related to bladder cancer patients, and a prognostic risk assessment nomogram was constructed, the accuracy of which was evaluated with calibration curves.@*RESULTS@#A prognostic risk scoring equation for bladder cancer patients was constructed based on nine cuproptosis-related lncRNAs. Immune infiltration analysis showed that the abundances of M0 macrophages, M1 macrophages, M2 macrophages, resting mast cells and neutrophils in the high-risk group were significantly higher than those in the low-risk group, while the abundances of CD8+ T cells, helper T cells, regulatory T cells and plasma cells in the low-risk group were significantly higher than those in the high-risk group (all P<0.05). Kaplan-Meier survival curve analysis showed that the total survival and progression-free survival of the low-risk group were longer than those of the high-risk group (both P<0.01). Univariate and multivariate Cox analysis showed that the risk score, age and tumor stage were independent factors for patient prognosis. The ROC curve analysis showed that the area under the curve (AUC) of the risk score in predicting 1, 3 and 5-year survival was 0.716, 0.697 and 0.717, respectively. When combined with age and tumor stage, the AUC for predicting 1-year prognosis increased to 0.725. The prognostic risk assessment nomogram for bladder cancer patients constructed based on patient age, tumor stage, and risk score had a prediction value that was consistent with the actual value.@*CONCLUSIONS@#A bladder cancer patient prognosis risk assessment model based on cuproptosis-related lncRNA has been successfully constructed in this study. The model can predict the prognosis of bladder cancer patients and their immune infiltration status, which may also provide a reference for tumor immunotherapy.

RESUMO

Objective To investigate the therapeutic effect of eperisone on patients with subacute spinal cord neuropathy with incomplete spasm. Methods Forty-eight patients with Subacute spinal cord neurosis combined degeneration of both lower extremity incomplete spasm in Department of Neurology,Zhejiang Quhua Hospital were selected and randomly divided into control group and study group. The control group were treated with folic acid and mecobalamin, the study group received more with eperisone, and 14days for one course, two groups were treated for two course.Ashworth spasm Scale, simple Fugl-Meyer score scale, improved Barthel index, bilateral H tibial nerve reflex status, clinical efficacy and complications were compared before and after the treatment. Results After treatment, levels of the flexion and extension of knee joint in 2 groups increased, and the scores of Ashworth spasticity scale decreased, scores of FMA and MBI increased, the latency of H reflex latency increased, levels of Hmax/Mmax decreased(P<0.05),compared with the control group, levels of the flexion and extension of knee joint in the study group were higher, and the scores of Ashworth spasticity scale were lower, scores of FMA and MBI were higher, the latency of H reflex latency were higher, levels of Hmax/Mmax were lower(P<0.05),the effective rate of the control group(66.67%)was lower than the study group(91.67%)(P<0.05),the non-performing rate in 2 groups had no difference(25.00%Vs 20.83%).Conclusion Eperisone has definite effect on patients with subacute spinal cord neuropathy with incomplete spasms.it can decrease muscle tension and spasticity and improve neurological function.

RESUMO

@#Objective To investigate the effects of electroacupuncture (EA) on expression of phosphatase and tensin homology deleted on chromosome ten (PTEN) protein and growth associated protein-43 (GAP-43) in rats with Barrel cortex focal ischemia. Methods 24 adult male Sprague-Dawley rats were divided into sham group (n=8), model group (n=8) and EA group (n=8). The EA group accepted EA at Baihui (DU20) and Fengfu (DU16) acupoints 3 days after modeling, 30 min a time, once a day. They were assessed with Corner Test 1 day before modeling, and 3 days, 7 days and 14 days after medication. The expression of GAP-43 and PTEN around infarct zone were detect with Western blotting 14 days after medication. Results The frequence of turn-right decreased in the EA group compared with that in the model group (P<0.001) 14 days after modeling. The expression of GAP-43 increased in the model group compared with that in the sham group (P<0.05), and increased more in the EA group than in the model group (P<0.05). There was no significantly difference in PTEN expression between the model group and the sham group (P=0.460), and significantly decreased in the EA group (P<0.001). Conclusion EA may inhibit expression of PTEN protein and increase expression of GAP-43, which may be involved in nerve regeneration and functional recovery.

RESUMO

BACKGROUND AND OBJECTIVEPatients with malignant tumor are at high risk of thrombophilia, which contributes to thromboembolism. Surgical treatment is one of the critical risk factors. In this study, changes and clinical significances of blood coagulation of lung cancer patients pre- and post operation were investigated.

METHODSA prospective, controlled study were carried out in 74 lung disease patients, who were divided into lung cancer group and benign lung disease group. In each group, pre- and postoperative changes in prothrombin time (PT), activated partial thromboplastin time (APT), platelet count (PLT), D-dimer (D-D) and fibrinogen (Fib) and clinical performances were observed and compared in intra- and intergroups.

RESULTSThe concentration of Fib both in lung cancer group and its subgroup (adenocarcinoma of lung) increased, preoperative differences between benign lung disease group and subgroup (squamous cell carcinoma of lung) was significant (P < 0.05). PT (postoperative 1st to 7th day) in lung cancer group prolonged, APTT (postoperative 3rd to 7th day) reduced, Fib (postoperative 3rd to 7th day) and D-D (postoperative 1st to 7th day) increased, PLT reduced on the 1st, 3rd day but then increased on the 5th, 7th day after operation, the difference between pre- and post-operation was significant (P < 0.05). D-D and PT in lung cancer group on the 7th day was longer than in benign lung disease group (P < 0.05). One pulmonary thromboembolism (PTE) case in lung cancer group occurred, while in benign lung disease group none venous thromboembolism (VTE) appeared.

CONCLUSIONPatients with lung cancer are in high hypercoagulable state, and prone to VTE. It is necessary to take some interventions to avoid VTE.
